New Study in Experimental Biology and Medicine Shows the Role of Sphingosine Kinase 2 as Therapeutic Target for Diabetes

A recently published article in Experimental Biology and Medicine (Volume 248, Issue 1, January 2023) provides a mechanism by which Sphingosine kinase 2 (ShK2) impacts Insulin receptor (InsR) trafficking, thereby regulating insulin signaling. The study, led by Dr. Pu Xia, in the Department of Endocrinology and Metabolism at the Zhongshan Hospital and Fudan Institute for Metabolic Diseases of Fudan University (China), describes that inhibition of ShK2 results in attenuation of clathrin mediated endocytosis of InsR.

Disturbed InsR trafficking is associated with impaired insulin signaling and the development of type 2 diabetes (T2D). Sphingosine kinase 2 (SphK2) is a lipid kinase that regulates a variety of cellular processes, including acting as a key regulator of hepatic insulin signaling and glucose homeostasis. However, the specific function of SphK2 in insulin receptor trafficking remained undefined leading to the current study by Aji et al.

This study, led by Dr. Xia, analyzed the role of Sphingosine kinase (SphK2) in regulating insulin receptor trafficking by analyzing Huh7 cells, which is an immortalized cell line derived from male hepatoma tissue, treated with a specific SphK2 small molecule inhibitor or siRNA. Huh7 cells are often used as a substitute for primary hepatocytes. Flow cytometry and immunofluorescence assays were used to investigate the role of SphK2 in insulin receptor trafficking. SphK2 was found to be able to regulate insulin receptor traffic, while SphK1, another isoform of Sphingosine kinase, did not. These findings demonstrate that SphK2 plays an essential function in controlling hepatic insulin signaling by regulating insulin receptor trafficking. 

Dr. Steven R. Goodman, Editor-in-Chief of Experimental Biology and Medicine, said, "Xia and colleagues have provided clear insight into the mechanism by which Sphingosine kinase 2 regulates Insulin Receptor endocytosis, and therefore Insulin signaling and glucose homeostasis. In doing so, they identify a new therapeutic target for treatment of Type 2 Diabetes."

Diabeloop is planning a dedicated study for people living with Type 2 diabetes to evaluate the efficacy and clinical benefits of the combined technologies.

Diabeloop accelerates its interoperability strategy with connected pens 

On the occasion of the annual congress of the Société francophone du diabète (SFD), in Montpellier, March 21-24, Diabeloop announces a collaboration with Novo Nordisk, a major global healthcare company.

The first milestone of the cooperation will focus on the integration of Novo Nordisk's new generation of connected pens, NovoPen® 6 and NovoPen Echo® Plus with DBL-4pen™.

DBL-4pen™ is a self-learning basal and bolus recommendation app for people with Type 1 and Type 2 diabetes who take multiple daily insulin injections (MDI). NovoPen® 6 and NovoPen Echo® Plus are connected pens with a dose memory function that keeps a history of the last 800 insulin injections and shows on a built-in digital display the number of units administered during the last injection, as well as the time since that injection. Data transfer from the connected pen to an external device is based on Near Field Communication (NFC). These connected pens are the only to be covered by public health insurance in France.

The common commitment of the two companies to bring innovative solutions for diabetes management to patients has driven the conclusion of this agreement.

Upcoming launch of a clinical program with Type 2 diabetes patients 

The clinical program will start with an initial multi-brand, multicentric before/after 14-week clinical trial that aims to assess the efficacy and safety of DBL-4pen™ mobile application on glycemic control in patients with Type 2 diabetes. It will include forty (40) patients with T2D across four (4) hospitals in France. 

Pr. Pierre-Yves Benhamou, Chief Medical Officer of Diabeloop, comments: "With this interventional, open, single-arm clinical trial involving adults with Type 2 diabetes, we aim to evaluate efficacy, safety and also treatment compliance. In addition, we will measure the improvement in quality of life and satisfaction of the enrolled patients and also evaluate the effect of DBL-4pen™ on the development of HbA1c in the extension phase."

Diabeloop will then initiate a dedicated clinical study, to assess the efficacy and the clinical benefits of the combined solution connecting DBL-4pen™ with NovoPen® 6 and NovoPen Echo® Plus connected pens. NovoPen® 6 and NovoPen Echo® Plus data will transmit to the DBL-4penTM self-learning algorithm which will recommend the dose of insulin to be injected in real-time from input data such as meals and exercise.

This trial program is shaping Diabeloop's future projects for MDI therapy. Diabeloop plans to address first the Type 2 population on intensive insulin treatment before soon expanding to people with Type 1 diabetes who do not have access to or desire to wear an insulin pump.

DBL-4pen™, self-learning basal and bolus recommendation app for pen users

Diabeloop has developed a unique smartphone app that embeds its self-learning algorithm to dynamically titrate basal and bolus insulin doses. The app is connected to a connected insulin pen and a continuous glucose monitor (CGM).

Directed by Melanie Orr from a screenplay by William Tyler Bell, Harm’s Way has the following synopsis: The proprietor of a women’s shelter jeopardizes the relationship between an abused mother and daughter. Darlene and her daughter Victoria seek refuge at a remote rural safe house for abused women that’s run by the strict and tough, but folksy and supportive Bea. Things turn sour when Bea proves to be a total lunatic with an especially dangerous influence on the impressionable Victoria.

Well, in a curious move from the marketing department at Warner Bros., a new TV spot for Shazam: Fury of the Gods showcases a pretty big cameo reveal. CBR reports on the TV spot spoiler and director David F. Sandberg’s reaction (although they also put the spoiler in the url, so tread carefully).

Amid the shake-ups and reinvention of the DC Film universe, with James Gunn and Peter Safran spearheading a ten-year plan, some of the existing franchise sequels are still scheduled for release, including Aquaman and the Lost Kingdom and The Flash. However, Shazam: Fury of the Gods is set to release later this month and promotion is already up and running. This includes at least one TV spot that spoils a cameo appearance that would have surely excited some of the fans and garner applause at the theater. Although, it still might, depending upon who has seen the commercial.

In one of the TV spots for the film, there is not only a shot but an extended exchange between Shazam and Wonder Woman, played once again by Gal Gadot, as she appears to help bail him out of a jam. This is an official release and not leaked clip. The director of the Shazam movies, David F. Sandberg, has addressed this on Twitter with a post saying, “Well there’s some big Shazam spoilers out there now. If you want to go in fresh maybe don’t be online or watch tv with ads… Pretty good advice in general actually.”

Zachary Levi, the film’s star, would quote the director’s tweet as he backs it with his post simply saying, “THISSSSSSS!!!” One of the big perks of having a shared universe is being able to crossover characters from one movie to the next, and these sequels usually play their cards when they can. What will particularly be a bummer for fans is this may be the last time we see Gal Gadot in the role after her four film appearances. Plans for a third Wonder Woman had fallen through as controversy around Patty Jenkins’ departure led to the project’s demise.

Shazam: Fury of the Gods is due to zap into theaters on March 17.

By the way, I must say in Sideshow Bob-like fashion, I’m well aware of the irony of posting the spoiler in reporting that the director advises fans to stay away from spoilers.

Dwayne Johnson weighs in on fighting to bring Henry Cavill back to the DCU, only to have his role cut by new studio management.

While cruising the red carpet in his sharp salmon-colored suit at last night’s Academy Awards celebration, Dwayne Johnson got real about Henry Cavill’s exit from the DCU. While smiling for the cameras, Variety‘s senior culture and events editor Marc Malkin asked Johnson about his feelings toward Cavill’s big comeback getting ditched by the studio’s new management.

“All that I can do, and all that we could do when we were making ‘Black Adam,’ was to put our best foot forward and surround ourselves with the best people and deliver the best movie we could,” Johnson said. “Our audience score was in the 90s. Critics took a couple shots, but that’s just the business of it.”

“It’s almost like when you have a pro football team and your quarterback wins championships and your head coach wins championships and then a new owner comes in and says, ‘Not my coach, not my quarterback. I’m going to go with somebody new.’”

One of the highlights of Black Adam involves Black Adam (Dwayne Johnson) being confronted by Superman (Henry Cavill), with the Man of Steel warning the anti-hero about stepping out of line. The moment suggests a clash between the two titans is forthcoming. However, with James Gunn and Peter Safran now in charge of the studio, the battle between the two omega-level punching bags has been postponed indefinitely.

It’s hard to imagine a world where Johnson isn’t bitter about Gunn and Safran taking DC Studios in a new direction. Johnson tried for 15 years to get Black Adam into theaters, promising fans a changing of the hierarchy at DC with the anti-hero’s arrival. Unfortunately, Black Adam underperformed at the box office, bringing the character’s future at the studio into question.

“After 15 years of relentless hard work to finally make ‘Black Adam,’ I’m very proud of the film we delivered for fans worldwide. I will always look back on the fan reaction to ‘Black Adam’ with tremendous gratitude, humility and love,” Johnson told fans last December.

Johnson insists he and James Gunn are in touch and that Black Adam has a future at the studio. “James Gunn and I connected, and Black Adam will not be in their first chapter of storytelling. However, DC and Seven Bucks have agreed to continue exploring the most valuable ways Black Adam can be utilized in future DC multiverse chapters.”

Clairemont Elementary School and John R. Lewis Legacy Institute Announce March on Decatur and Conversation of Understanding

After the overwhelming success of the March on Clairemont during the 2022-2023 school year, Clairemont Elementary School committed to expanding efforts to include the entire Decatur community. It is with extreme excitement that the budding equity and social justice leaders at Clairemont Elementary School present March on Decatur.  On Friday, March 31, 2023, at 9:30 AM, Clairemont Elementary will lead a march from 155 Erie Avenue that will culminate on the stairs of the Decatur History Center, where those gathered will hear from dynamic community leaders and student activists. The theme for the march is "Extinguishing the Isms."

"We are confident that support from our school community can and will bring attention to and boldly take steps to eliminate sexism, heterosexism, ableism, and racism," said Clairemont Elementary principal Dr. Curtis Armour. In the month leading up to the march, students will participate in an age-appropriate curriculum that focuses on each of these "isms." At the conclusion of the experience, students will choose the social justice issue that is most meaningful to them and create chants, posters and banners that demand change.  

Also, The Clairemont Elementary School Equity Team, PTA/DEI Committee, and the John R. Lewis Legacy Institute will host a Conversation of Understanding, an initiative to raise awareness, educate, and unite the Clairemont community. The event will take place on Wednesday, April 12, 2023, from 5:30 PM- 7 PM in the Clairemont Auditorium. The moderators for the evening will be Mr. Garry Lowe and Mrs. Chanell Huff-Cox.

The Conversation Champions (Panelists) will tackle the rich and complex topic of diversity and inclusion by bravely sharing challenging personal experiences. They will highlight biases and stereotypes they have encountered, and how these difficult situations impacted them.  Speakers will also share practical tips on how we all can act in a more inclusive manner. "We will also emphasize allyship among our residents. More specifically, identifying opportunities to be an effective ally, explaining what allyship looks like on a day-to-day basis and highlighting the positive impact that allies can have on under-represented communities," said JRLLI Executive Director Jerrick Lewis.  

The John R. Lewis Legacy Institute, Inc. (JRLLI) is a nonprofit organization comprised of members of Congressmen Lewis' immediate family, whose mission is to promote diversity, support civil rights education, and engage in community service projects, including but not limited to Social Justice, Equity in Education, and Health Awareness, enabling future generations to continue the legacy of good and necessary trouble.
